Cancer susceptibility

Cancer susceptibility gene mutation term used to describe mutations (changes) in certain genes that may increase a person’s risk of some types of cancer. For example, a person who has certain mutations in the BRCA1 or BRCA2 gene has higher than normal risks of developing breast cancer and ovarian cancer. Cancer susceptibility gene mutations are usually inherited (passed from parent to child) and may be seen within families. Knowing if a person has a cancer susceptibility gene mutation may help prevent, diagnose, and treat cancer. Not all people who have a cancer susceptibility gene mutation will develop cancer. Also called cancer predisposing gene mutation. Cancer susceptibility is due to interactions between inherited genetic factors and exposure to environmental carcinogens. The genetic component is constituted mainly by weakly acting low-penetrance genetic variants that interact among themselves, as well as with the environment.
The identification of human tumor susceptibility genes and discovery of their roles in carcinogenesis will ultimately be important for the development of methods for prediction of risk, diagnosis, prevention and therapy for human cancers. Recent studies show that low-penetrance mutant alleles of some tumor suppressor genes, as well as the better known high-penetrance mutations, can contribute to cancer incidence. These low-penetrance alleles confer a significantly increased risk of sporadic tumors. We suspect that a family may have inherited a mutation in a cancer susceptibility gene when we see one or more of the following:
1. Several people in the family with similar or related cancers
2. Cancer affecting more than one generation on the same side of the family
3. Cancer that starts at an earlier age than average (often in the 30s or 40s)
4. Individuals who have had more than one type of cancer (for example, both breast and ovarian cancer, or cancer in both breasts)
Cancer susceptibility genes have been identified for many different types of cancer, including:
1. Breast
2. Ovarian
3. Colon
4. Uterine
5. Kidney
6. Medullary thyroid
